Our laboratory aims to unravel pathogenesis induced by important zoonotic pathogens and develop intervention strategies. We established the steller role of virulence-plasmid in Aeromonas hydrophila-pathogenesis and successfully used plasmid-cured isolates as vaccine candidates against its spread. MDR-A. hydrophila is global issue. We sequenced Ah-virulence-plasmid, demonstrating quinolone and beta-lactamase resistance is plasmid-encoded, opening new avenues in controlling the bacterium. Our group reported the role of calcium and its dependent kinases in macrophage apoptosis induced by zoonotic pathogens, and established the importance of ER-mitochondrial Ca-dynamics in the process. We demonstrated how zoonotic-pathogens exploit TLR4-topology for immune-evasion and persistence inside the host. The strikingly similar mechanisms of cell death induced in fish and mammals clearly suggested that pathogenic mechanisms employed by zoonotic-pathogens transcend species barrier. Toxoplasma gondii resides perpetually inside infected host. Work emanating from our laboratory described how the parasite manipulates β-catenin signaling and usurps tryptophan-metabolites for intracellular survival, suggesting alternate approach for its treatment.

Xenobiotic-immunotoxicity has been the other major focus of my group. We mapped the course of events leading to profound immunosuppression caused by arsenic and identifed the signature genes involved in arsenic-induced apoptosis.These findings are important from the viewpoint of predictive toxicology and biomarker development. Recently, we elucidated that chronic endosulfan-exposure cause immune commotion wherein the immune-organs per se, generate endosulfan-metabolites, principal components of endosulfan-toxicity, thereby compounding the toxic effects of the pesticide. Fluorosis is a major health issue and our work on fluoride-induced immunotoxicity has contributed significantly towards the wellness of the society.
